High resolution whole genome profiling by array

The Netherlands Array-based comparative genomic hybridization (arrayCGH) is a high resolution screening method for the detection of DNA copy number aberrations. Through the selection of thousands of large-insert clones such as BACs throughout the genome this copy number screening can be performed on a genome-wide scale (molecular karyotyping). In Nijmegen we have collected a genome-wide set, cultured the BACs, isolated DNA and amplified this DNA by means of DOP-PCR. In this presentation the production of these arrays will be outlined and our first results using arrays consisting of approximately 3500 clones will be presented. Our results show the potential of this approach for whole-genome copy number screening.
